본 연구에서는 천연식물 유래 방향성 에센셜 오일의 항천식 효과 및 항염 효과에 관한 연구를 수행하였다. 알레르기성 천식은 기도 염증, 점액 과분비 및 기도 과민반응이 나타나는 호흡기 점막의 면역 질환이다. 기도 염증은 T세포, B세포, 비만세포, 호중구 및 호산구 등의 염증 세포들과 사이토카인, 케모카인과 같은 화학매개체에 의해 유발되며 기도과민성, 폐의 염증 및 혈청 내 IgE 분비 증가 등이 대표적인 특징으로 나타난다. 알레르기성 천식을 유발한 생쥐모델을 이용하여 대조군과 실험군의 염증억제효과 및 그 기전을 규명하는 연구를 수행하였다. Ovabumin(OVA)으로 알레르기성 천식을 유발시킨 천식 생쥐 모델을 사용하여 실험군에 0.025% 로즈마리, 마조람, 사이프러스 혼합물(RTC-mixture)을 8주간, 1주 3회, 1회 30분간 흡입시켰다. 실험일로부터 총 12주 후에 살처분시켜 유세포 형광 분석기로 폐세포 내에 존재하는 면역세포 비율을 분석한 결과 대조군에 비해 RTC-mixture 흡입군에서 granulocytes/lymphocytes, CD3e+/CCR3+, CD4+/CD8+, CD69+/CD3e+, IgE+/B220+ 및CD11b+/Gr-1+의 세포비율이 감소하였다. 결론적으로 알레르기성 천식모델 생쥐에서 RTC-mixture 흡입군은 폐세포 내 granulocytes 감소, 호산구수 감소, T 세포 감소, IgE 생성량 감소, 호산구의 경내피로의 이동억제에 관여함으로써 천식으로 인한 알레르기성 염증 반응을 억제시키는데 영향을 주는 것으로 사료되었다.In this study, we tried to evaluate the anti-asthmatic and anti-inflammation effect of natural plant derived aromatic essential oil. Allergic asthma is a kind of immunologic disorder in respiratory epithelium, in which inflammation of airway, hypersecretion of mucus, and hypersensitivity of airway are observed. Allergic inflammation is induced by chemical mediators derived from inflammatory immune cells including T cells, B cells, mast cells and eosinophils, and chemical mediators such as cytokine or chemokine, characterized by pulmonary inflammation and increase of serum IgE level etc. To evaluate and compare the inflammatory mechanism between control group and experimental group, mouse allergic asthma model was selected.
0.025% of rosemary, marjoram, and cypress mixture (RTC-mixture) was nebulized to asthma induced mice for 8 weeks on a basis of 3 times per week and 30 min of nebulization each time. After 12 weeks from the initial experiment, the mice lungs were removed and analyzed by flow cytometer, and expressed as percentages in granulocytes/lymphocytes, CD3e+/CCR3+, CD4+/CD8+, CD69+/ CD3e+, IgE+/B220+ and CD11b+/Gr-1+cells. In the RTC-mixture treatments group, the percentages of granulocytes, CD3e+/CCR3+, CD4+/CD8+, CD69+/CD3e+, IgE+/B220+ and CD11b+/Gr-1+cells were decreased when compared with control group. In conclusion, RTC-mixture have profound effects on allergic inflammation in a murine model of asthma through suppression of granulocytes, eosinophils CCR3 expression, IgE, and CD11b cells expression.
