과거 의복 재료로 널리 사용되어 왔던 실크는 단백질 섬유 자체로써의 물리적 성질 및 화학적 성질이 매우 우수할 뿐 만 아니라 그 주요 구성 성분인 글리신, 알라닌, 세린 등이 보습력, 자외선 차단력 등에서 뛰어난 효능을 지니는 등 피부와 매우 친화적인 성분이라는 점에서 피부 보습제 등 기능성 미용 재료의 하나로 최근 주목받고 있다.
본 연구에서는 이러한 실크의 기능적인 면을 고려한 모발 화장품으로써의 이용 가능성을 타진하고자 누에고치 가수분해물인 저분자 실크 펩타이드 (Silk peptide)를 포함하는 모발 화장품을 개발하고, 이에 대한 헤어 컨디셔닝 효과를 평가하였다. 그리고 헤어 컨디셔닝 제품 제조 시, 실크 펩타이드를 분자량과 성분 함량의 차이를 두어 다양한 헤어 컨디셔너 (Hair conditioner)를 제조하고, 이에 대한 컨디셔닝 효과를 비교하고자 하였다.
각각의 실크 펩타이드 컨디셔너 (Silk peptide conditioner) 샘플에 대한 헤어 컨디셔닝 효과는 질량, 인장강도, 흡광도, 보습력 등 기초적인 물성 평가를 통해 분석함과 동시에 나아가 모발의 미세 구조적 변화를 Atomic Force Microscopy (AFM)을 이용하여 분석하였다.
그 결과 실크 펩타이드의 성분 함량에 따라 약간의 제품 효능 차이는 존재하였으나 전반적으로 실크 펩타이드 컨디셔너 처리 후의 모발 샘플들은 손상모와 대조군 (control)과 비교하여 모발의 질량 및 굵기, 그리고 보습 효과 증진 등 기초 물성에 있어 향상된 수치를 나타내었으며, 미세 구조적으로도 모발 표면의 손상 정도와 모발의 두께, 큐티클의 height 변화에 있어 건강모와 같은 수준으로 회복하였다.
특히 AFM의 line profile을 통한 미세 구조 분석 결과, 실크 펩타이드를 원료로 한 헤어 컨디셔너 조성물이 화학시술로 인하여 손상된 모발 표면에 흡착, 큐티클 손상부를 메워주는 것이 확인되었다.
따라서 실크 펩타이드를 원료로 한 헤어 컨디셔너는 향후 효과적인 천연 헤어 컨디셔닝 제품의 하나로 적용될 수 있을 것으로 사료된다. 아울러 방법론에 있어서도 본 연구에서 제시한 AFM을 이용한 모발의 미세구조 분석법은 모발의 표면에 대한roughness 등 다양한 정보를 나노 단위의 height 등 다양한 물리적 수치로 제시할 수 있을 뿐 만 아니라 3차원적 이미지로 나타낼 수 있다는 점에서 기존의 2차원 평면 주사전자현미경 (Scanning Electron Microscope: SEM)을 통한 모발의 미세구조 분석법의 한계를 극복할 수 있을 것으로 기대된다.
