본 연구에서는 바이오알코올의 제조과정에서 당화에 필요한 Agarivorans albus YKW-34 유래 재조합 β-agarase B(AgaB34)에 대한 생화학적 특성 및 응용에 대해 연구를 진행하였다. 재조합 AgaB34 를 대장균(Escherichia coli)에서의 발현양을 높이기 위하여 코돈을 최적화하였다. 또한 발현 유도제인 IPTG 의 첨가시간 및 농도에 대한 최적 조건을 탐색한 결과, 18% 향상된 결과를 나타내었다. 재조합 AgaB34 가 발현된 추출물(crude extracts)에 대해 황산암모늄 침전법과 DEAE-Sepharose 컬럼크로마토 그래피를 이용하여 재조합 AgaB34 를 정제한 결과, 약 14 배의 정제도와 59 %의 수율을 나타냈다. 재조함 AgaB34 에 대한 특성화 결과 아가로오스에 대한 Km, Vmax, kcat, kcat / Km 값은 각각 0.0238 mM, 49.623 μmol/mg/min, 40.526 /s, 1703.919 /s·mM 로 나타났으며, 최적 pH 는 7.0, 최적온도는 40 ℃ 를 나타냈으며, pH 5-9 에서 1 시간 및 50 ℃ 에서 30 분 보관 후에도 활성의 80% 이상을 유지하고 있었다. 또한, Cu2+, Mn2+, Zn2+ 및 Al3+ 이온이 AgaB34 활성을 강하게 저해하였다.
한편, 새로운 분석법인 ML 법을 개발한 결과 기존 30 분이던 활성측정시간을 5 분으로 줄일 수 있었으며, 아가로오스 분해속도를 측정할 수 있는 효소활성 측정법이 개발되었다. 그러나, 활성측정결과의 정밀성이 9%내외로 높게 나타나 향후 이 문제를 보완한다면 기존 β-agarase 활성측정법을 대체할 수 있을 것으로 보인다.
AgaB34 의 효소반응 메커니즘을 연구하고자 기질 결합부위로 알려진 틈새(cleft)구조의 기질결합 잔기에 대한 변이체를 부위특이적 변이법을 이용하여 만들어 변이체들에 대한 생화학적 특성을 조사하였다. 그 결과, 틈새(cleft)구조의 기질결합 잔기의 변이가 효소활성에 영향을 미쳤으며, AgaB34 의 활성이 다른 고활성을 갖는 β-agarase 보다 낮은 원인이 이들 부위의 치환에 따른 것임을 확인할 수 있었다. 또한, T59K 및 T261I 변이체에 대한 열안정성의 변화를 확인한 결과 열안정성이 5 ℃ 가량 상승 되었음을 확인할 수 있었다.
효소의 산업적 응용성을 높이기 위하여 최적의 고정화 방법을 탐색하였다. AgaB34 에 대해 포괄법(entrapment), 물리적 흡착법, 이온결합, 공유결합 및 가교법(가교화된 효소를 포괄법으로 고정)에 의한 고정화를 수행하고 이들의 특성화를 실시한 결과, 열안정성은 가교법 및 공유결합법이 가장 우수해 20 % 향상되었으며, pH 안정성은 가교법이 가장 우수한 40% 높게 나타났으며, 재활용성은 공유결합법이 3 회 반복 실험에도 50% 이상 활성을 유지하여 가장 우수하게 나타났으며, 효소반응의 최적온도는 가교법이 60 ℃ 로 가장 높게 나타났다. 따라서, 가교법이 가장 우수한 고정화방법임을 확인하였다.
