메탄(CH₄)은 이산화탄소 다음으로 중요한 온실화가스이다. 지구에서의 메탄순화를 이해하기 위해, 본 연구에서는 두 가지 주요 메탄 방출 원인 반추위와 논토양에서 메탄생성균 및 산화균의 특징을 조사하였다.
한우 육성우(HGS), 한우 비육우(HFS), 젖소(HDC)와 염소(KNG)에서의 반추위 내 미생물 군집과 대사체분석을 위해 총세균 및 고세균의 16S rRNA 유전자의 바코드-파이로시퀀싱과 ¹H-NMR 기법을 각각 사용하였다. 대사체 및 반추위 미생물 군집을 이용한 통계적 비교는 나이 및 식이가 다른 HFS 와 HGS 에서는 유의한 차이가 없음을 나타내었다. 총세균 중 Bacteroidetes, Firmicutes, Fibrobacteres 와 Proteobacteria 문이 모든 반추위에서 우점하고, Fibrobacteres 문만이 KNG 에서 매우 낮은 양(<0.1%)이 있음을 생물정보학분석을 통해 확인하였다. 고세균 군집분석결과 거의 모든 염기서열들(sequencing reads)이 기존에 배양된 메탄생성균과는 떨어져있지만 연관은 되어있는 것으로 분석되었다. KNG 의 대사체 및 미생물군집은 다른 반추동물들과는 분명히 분리되는 것으로 통계분석을 통해 확인하였다. 추가적으로 HGS 와 HDC 의 세균 군집 및 대사체 프로파일이 비슷한 식이를 섭취했음에도 서로 다른것으로 조사되었다. 본 연구에서는 반추동물이 섭취하는 식이보다 반추동물의 종이 반추위에서 미생물군집과 대사체 프로파일을 결정하는 핵심요소임을 확인하였다.
논토양에서의 메탄방출량을 메탄생성균 및 산화균과 토양의 화학적특성과 함께 조사하였다. 메탄방출량은 모내기 후 증가하였고(7.2 mg/일∙㎡부터 552 mg/일∙㎡), pmoA 와 mcrA 유전자 발현량, mcrA 발현량/유전자 비, 온도 및 총유기탄소와 긍정적이고 유의성있는 상관관계를 보인 반면, 황산농도와는 부정적 상관관계를 나타냈다. 메탄산화균은 총세균의 16S rRNA 유전자염기서열들 중 단 0.79–1.75%로 작은 비율을 차지하는것으로 조사되었다. 유형II 메탄산화균인 Methylocystis 는 모내기 후 매우 빠르게 감소한 반면 Methylosinus 와 미분류된 Methylocystaceae(유형 II)는 벼재배기간동안 비교적 일정하게 유지하는 것으로 나타났다. Methylocaldum, Methylobacter, Methylomonas 와 Methylosarcina (유형 I)는 초기기간동안은 매우 적은 비로 존재하다가, 60 일 이후부터 증가하여서 최대존재비는 90–120 일에 관찰되었다. 33,318 개의 고세균 염기서열들 중 68.3–86.6%가 메탄생성균으로 분류되었고, 메탄생성균들 중 Methanosaeta, Methanocella, Methanosarcina 와 Methanobacterium 이 우점하는 것으로 나타났고, 최대존재비로 60–90 일에 관찰되었다. 단 4 개의 염기서열들만이 혐기적 메탄산화균으로 분류되어, 혐기적 메탄대사는 논토양에서는 무시할 수 있음을 시사하였다. 결과들을 통합하여 다변량 정준상관분석을 수행한 결과 정규화된 mcrA/pmoA 유전자 발현량의 비율은 논토양에서의 메탄방출을 예측할 수 있는 유망 매개 변수로 활용할 수 있음을 찾을 수 있었다.
논토양에서의 총세균 및 고세균을 산소, 메탄과 총유기탄소의 농도와 함께 깊이에 따라 조사하였다. 비근권과 근권에서의 메탄, 총유기탄소 농도 및 미생물의 다양성은 깊이에 따라서는 비슷하게 조사되었지만, 메탄, 총유기탄소 농도 및 16S rRNA 유전자 수는 비근권보다 근권에서 더 높게 나타났다. 산소농도는 표면에서부터 줄어들어 8 ㎜ 에서 검출한계점 아래로 조사되었다. 16S rRNA 유전자의 파이로시퀀싱을 수행한 결과 세균과 고세균의 군집이 호기, 호기-혐기과 혐기영역에 따라 서로 다른것으로 나타났다. 호기적 메탄산화균은 호기-혐기 영역 부근에서 가장 많이 나타난 반면, 메탄, 총유기탄소 및 메탄생성균은 근권의 30–200 ㎜ 깊이에서 가장 높은것으로 조사되어, 메탄은 벼로부터 기인된 유기탄소를 이용하여 생성되고, 호기적조건에서 대사된다는 것을 제시하였다. Methylococcus, Methylomonas 와 Methylocaldum 과 같은 유형 I 메탄산화균의 상대적 빈도는 유형 II 메탄산화균(Methylocystis, Methylosinus)에 비해 깊이가 깊어짐에 따라 크게 감소하는것으로 나타났다. 메탄생성균 중 Methanosaeta 와 Methanoregula 가 모든깊이에서 우점하였고, Methanosaeta, Methanoregula, Methanosphaerula 및 GOM_Arc_I 은 깊이에 따라 증가하였다. 근권과 비근권의 깊이에 따른 (특히 표면에서의 밀리미터 규모) 메탄생성 및 산화균의 절대존재비의 대조를 통해 메탄생성균중 Methanosaeta, Methanoregula, Methanocella, Methanobacterium 및 Methanosphaerula 이, 메탄산화균중 Methylosarcina, Methylococcus, Methylosinus 와 미분류된 Methylocystaceae 가 in situ 에서 생리학적 활성을 가지는 것으로 규명하였다.
