시아노박테리아를 이용한 대사공학 및 합성생물학 기술은 이산화탄소의 탄화수소화학물질로의 직접적인 전환을 가능케 함으로써 대체 에너지 바이오연료의 생산이 가능하기에 주목을 받고 있다. 스쿠알렌은 불포화 탄화수소 (C30H50)로서 인체의 여러 조직에도 존재하며, 항산화 제품 및 의약, 화장품, 건강보조식품, 백신 보조제 등으로 이용되고 있다. 스쿠알렌 세계시장은 2021년에 이르면 총 2억 1,1170만 달러 규모에 도달할 수 있을 것으로 추정되며, 연평균 10.2 %의 성장세를 보일 전망이다. 그러나 상어의 간이나 식물성 오일로부터 유래한 스쿠알렌은 동물보호법 위배와 추출 운반 처리의 애로사항이 있다. 그래서 최근 미생물유래 스쿠알렌의 생산이 각광을 받고 있다. 특히 효모에서 생산하는 것은 포도당 기반으로 배양하여 대량으로 생산 시에 경제성이 떨어질 수 있다. 따라서 이산화탄소 광 전환이 가능한 시아노박테아를 통한 스쿠알렌의 생산은 많은 장점을 가진다.
본 연구에서는 대장균 균주 유래 외부유전자를 도입하여 시아노박테리아 대사경로를 수정하고, 스쿠알렌 생산 가능한 효모의 효소를 유전자를 S. elongatus PCC7942 균주에 도입하여, 스쿠알렌을 명반응, 고농도 이산화탄소 호기성조건에서 최대 생산하는데 최초로 성공하였다. 여기서 생산성을 더욱 높이기 위해 퓨전된 단백질을 제작하여 스쿠알렌 생산량이 증가하는 것을 확인하였다. 마지막으로 시아노박테리아의 대량배양의 적용을 위한 최적 생산조건을 맞춘 광반응기를 도입하여 시아노박테리아로부터 스쿠알렌을 생산하는 테스트를 진행하였고, 스쿠알렌을 명반응, 고농도 이산화탄소 호기성 조건 아래 생산하였다.
본 연구는 시아노박테리아를 이용하여 대사공학적 접근으로 명반응 고농도 이산화탄소 호기조건으로부터 스쿠알렌을 생산하였다. 더 나아가 스쿠알렌의 향상된 생산량을 갖는 우량 균주를 개발하였다. 더욱이 시아노박테리아에 대한 질소영양소 결핍의 극한 상태에서 유전자 발현의 정도를 대사흐름의 관점에서 모니터링 함으로써 향후 유전자 재조합과 시아노박테리아 연구에 기반이 될 수 있는 연구도 병행하였다.
