Due to the development of biodegradable material and cell culture technique, cultured skin substitutes

have been diversified with variable matrices and cells. Since the hair follicles contain the stem cells for not only hair

follicles but also skin structure, various hair follicle-derived cells have been tried for the skin substitutes. Dermal

sheath(DS) cells were used for dermal fibroblasts and outer root sheath(ORS) cells were replaced with epidermal

keratinocytes. Seeded chitosan sponges and collagen gels with DS and ORS cells were tested for skin substitutes in

full-thickness wound of rat dorsal skin. In result, both chitosan sponges and collagen gels with DS cells performed

well as dermal supporting matrices for cell migration and proliferation in wound healing, while the collagen gels

with ORS cells helped the better epidermal regeneration than the sponges. Especially, the combination of chitosan

sponges and collagen gels seeded with DS and ORS cells, respectively regenerated well epidermal and dermal

reconstruction.
