제1세부과제명 : 벼 과민성인산화조절 핵심유전자, OsEDRI 이용 병저항성 벼 육성
Mitogen-activated protein kinase (MAPK) cascade(s) is important signalling pathway for plant defense/stress responses. Though MAPKs have been identified and characterized in rice (Oryza sativa L.), a monocot cereal crop research model, the first upstream component of the kinase cascade, namely MAPK kinase kinase (MAPKKK) has not yet been identified. Here we applied its hypersensitive regulating function on developing resistant rice. We made promoter change through insertion of multiple WRKY binding W-boxes in the OsEDR1 promoter. The W-box is known to regulate defense-related genes induced by biotic stresses. Enhanced transcription of PR-genes were screened with RT-PCR when transgenic rice lines were tested. Some disease resistance was also detected when transgenic rices were applied with rice blast fungus M. oryzae. These results suggest a role for OsEDR1 in defense/stress signalling pathways and its application for generation of disease resistant transgenic rice cultivar.
제2세부과제명 : N-O-m를 이용한 파이토알렉신 과생산벼 형질전환체 육성
Phytoalexins have antipathogenic activities including antibacterial or antifungal activityies. Rice produces phytoalexin, sakuranetin whose structure is 7-O-methylnaringenin. Sakuranetin has been known to inhibit growth of Magnaporthe grisea. It is assumed that sakuranetin is synthesized from naringenin via one methyltransferase reaction. A O-methyltransferase (OMT) from Streptomyces avermitilis was cloned. This OMT was shown to transfer a methyl group to 7-hydroxy group of naringenin. Thus, it was naringenin 7-O-methyltranferase. Using this OMT, rice expression vector was constructed. Rice transformation was carried out. Rice transformants expressing naringein 7-O-methyltransferase were selected. The expression of naringenin 7-O-methyltransferase was confirmed using RNA blot. Anlaysis of total phenolic compounds of transformant using high perfermance liquid chromatography showed that sakurantenin was produced. Thus, overexpression of narigenin 7-O-methyltransferase in rice could establish the production of sakuranetin.
