이전 연구에서 우리는 세균의 철분 킬레이트제 성분인 데프록사민(deferoxamine, DFO)이 염증신호로서 작용을 하여 사람 장 상피세포에서 IL-8 케모카인 생성을 유도하고, 이는 ERK1/2 및 p38 MAPK 인산화 효소의 활성으로 매개되고, NF-κB에 비의존적임을 보고하였다. 본 연구에서는 novel protein kinase C (PKC) 이성질 체인 PKCδ가 철분 결핍에 의한 IL-8 생성에 필요하다는 것을 발견하였다. PKCδ 저해제인rottlerin을 세포에 처리하면 데프록사민이 유도한 IL-8 생성이 저해된 반면, conventional PKC 이성질체의 저해제인 Gö6976은 저해효과를 유의하게 보이지 않았다. 상피세포의 PKCδ는 데프록사민에 의해 빠르고 강하게 인산화되었다. PKCδ를 과발현시켰을 때 데프록사민에 의한 인산화가 강하게 일어난 반면, PKCδ의 인산화 효소 불활성형 돌연변이 DNA를 도입하였을 때 PKCδ의 인산화가 억제되었다. 특이적인 siRNA를 사용하여 세포내재성 PKCδ 단백질의 수준을 감소시켰을 때도 데프록사민에 의한 IL-8 생성이 유의하게 저해되었다. 이러한 결과들을 통해, 사람 장상피세포에서 철분 결핍으로 유도한 IL-8 생성을 조절하는 데 있어 PKCδ가 중요한 역할을 하는 것을 알 수 있었다.Our previous study demonstrated that a bacterial siderophore, deferoxamine (DFO), could trigger inflammatory signals in human intestinal epithelial cells as a single stimulus, leading to IL-8 production via ERK1/2 and p38 phosphorylation and NF-κB-independent mechanism. In the present study, we proved that a novel protein kinase C (PKC) isoform, PKCδ, is necessary for DFO-induced IL-8 production. Pretreatment of HT-29 cells with rottlerin showed remarkable inhibition of DFO-induced IL-8 production. In contrast, a conventional PKC inhibitor Gö6976 did not show significant inhibition of DFO-induced IL-8 production. DFO induced strong phosphorylation of PKCδ in the epithelial cells. Overexpression of PKCδ resulted in enhanced PKCδ phosphorylation, while transfection with dominant-negative PKCδ vector failed DFO-induced phosphorylation. In addition, transfection of HT-29 cells with siRNA targeting endogenous PKCδ, which suppressed PKCδ expression, attenuated DFO-induced IL-8 production. These results demonstrate that PKCδ plays an important role in regulating iron chelator-induced IL-8 production in human intestinal epithelial cells.
