배경 및 목적:양적 형질에 대한 유전적 관련성 분석을 할 경우, 유전자간의 복잡한 교호작용과 유전자와 환경적 요인간의 교호작용 등을 고려할 필요가 있는데, 만약 단일 염기 다형성의 수가 많아지면 그것들 사이의 복잡한 구조에 의한 영향을 파악하는 데에는 분석 방법 자체에 어려움이 발생할 수 있다.이에 본 연구에서는 이를 효율적으로 극복할 수 있는 분석 방법으로 혼합모형을 이용한 방법을 제안한다. 또한 본 연구에서는 실제 4개의 단일 염기 다형성들을 이용하여 혈중 지질농도에 대한 유전자들 간의 고차원적인 교호작용의 유의성을 평가한다.방 법:본 연구에서는 혼합모형에서 고정 효과로서 연령, 성별, 체질량지수, 흡연력, 음주력, 운동 여부 등의 환경적 요인들을고려하였고, 확률적 효과로서 유전형 집단 및 유전형 집단과성별과의 교호작용을 포함시켰다.단일 염기 다형성으로서는 AGT5, APOB, CETP3과 ACE6의 4가지를 이용했고, 혈중 지질농도들로서 총 콜레스테롤,중성지방, LDL-콜레스테롤, HDL-콜레스테롤의 수치를 이용하였다. 심혈관계질환에 대한 위험인자를 가지고 있지 않은남자 283명(42.1%), 여자 389명(57.9%), 총 672명의 건강한정상 성인들을 대상으로 분석하였다.
결 과:
혼합모형을 이용한 분석에서 총 콜레스테롤의 경우, 포괄
적인 유전형 집단의 효과가 유의하게 나타났으며(p=0.024),
성별과의 교호작용에 대한 효과 또한 유의하게 나타났다(p=
0.036). 중성지방의 경우에는 교호작용의 효과가 유의하게
나타났고, HDL-콜레스테롤에서는 유전형 집단의 효과가 유
의하게 존재하고 있었으며(p=0.043), 교호작용에 대한 효과
도 유의하게 나타났다(p=0.037). 구체적으로 어떠한 조합의
유전형 집단에 의한 효과가 유의하게 나타나는지 그리고 그
러한 효과가 성별에 따른 차이를 보이는 지를 개별적으로 평
가하였다. 총 콜레스테롤에서는 (CC, CC, AA, ID), (CC, CC,
GG, ID), (CT, CC, GG, II)의 경우 남자에서 더 낮은 것과
관련성을 보이고 있었다. 중성지방에서는 (CC, CC, AA, ID),
(CC, CC, GA, II), (CC, CT, GG, ID), (CC, CT, GG, II)들에
서 남자에서 더 낮은 것과 관련성을 보이고 있었으며, (CC,
CC, AA, II), (CT, CT, AA, ID) 및 (CT, CT, GG, ID)에서는
그와 반대의 관련성이 유의하게 존재하고 있었다. HDL-콜
레스테롤에서는 (CC, CC, GG, II)와 (CT, CC, GA, ID)에서
포괄적인 유전형 집단의 효과와 성별과의 교호작용이 유의하
게 존재했다.
결 론:
본 연구에서는 혈중 지질농도와 같은 양적 형질에서 관
찰되는 변동량이 다중의 단일 염기 다형성으로 구성된 유전
형 집단, 혹은 환경적 요인과의 교호작용 등에 의하여 영향
을 받는지 확인하기 위한 통계학적 방법으로 혼합모형을 이
용했다. 혈중 지질농도에서 각 유전형 집단들은 성별과의 유
의한 교호작용이 존재하고 있었고 그 양상은 유전형 집단별
로 각기 다르게 나타나고 있었다.Background and Objectives:Analyzing the association between multiple SNPs and the disease outcomes willprovide new insight into the disease’s etiology. However, this presents an analytic difficulty due to the largenumber of SNPs and the complex relationships among them. We proposed using the mixed model approach toidentify the significant multi-locus genotypes and the high-order gene-to-gene interactions. Subjects and Methods:We described the mixed effects model and applied this approach to real world data. For the purpose of theseanalyses, we examine the association of four types of SNPs (AGT5, APOB, CETP3 and ACE6) with the lipid profilesand the measures related with cardiovascular disease. We used data from 672 healthy individuals (283 males and389 females) who were without cardiovascular diseases. Results:The results of our analysis suggested that therewere significant random genotype patterns and genotype groups according to the gender effect on the lipid profiles.In other words, there was significant variability across the genotype groups because of the effect of gender on thelipid profiles. Conclusion:The mixed model approach provided a flexible statistical framework for controllingpotential confounding variables and for identifying a significant genetic contributions that may come aboutthrough the effects of multi-locus genotypes or through an interaction between the genotype and environmentalvariables (e.g. gender) with the variations in quantitative traits (e.g. lipid profiles). There were significant geneticcontributions to the variability in the lipid profiles, and these were explained by the 4 SNPs described in our realdata. (Korean Circulation J 2006;36:229-235)
