Eph 수용체와 Ephrin 리간드는 척추동물의 배아발생 과정에 있어 광범위한 부분에서 발현된다. Eph
수용체와 Ephrin 리간드의 상호 보완적인 발현을 통해 Eph family는 신경계의 형성 및 분화에 관련된 고유한 역
할을 수행할 수 있게 된다. 척추동물의 등쪽 중뇌 (dorsal mesencephalon)는 크게 앞부분(anterior)과 뒷부분
(posterior)으로 나뉘게 되는데, 이곳의 Eph 수용체와 Ephrin 리간드의 상호 배타적인 발현은 중뇌와 주변의 간뇌
및 후뇌와의 경계선을 이루게 된다. 따라서 중뇌의 발생 기작을 연구하는데 있어서 Eph 수용체와 Ephrin 리간드
의 특이적인 발현이 어떻게 일어나는지를 밝혀내는 일은 아주 중요한 문제이다. EphrinA2와 EphrinA5는 중뇌
의 뒷부분(posterior mesencephalon)에서 발현되는 것으로 알려져 있고, 그것의 수용체인 EphA8은 중뇌의 앞부
분 (anterior mesencephalon)에서 발현되는 것으로 알려져 있다. 본 연구에서는 EphrinA2를 중뇌의 앞부분
(anterior mesencephalon)에서 과잉 발현하도록 유도한 형질전환 마우스를 제작하여 중뇌 발생 양상에 관해 연구
하였다. 그 결과, 중뇌의 앞부분에 해당하는 머리쪽 주름(cephalic fold)의 맞물림 부분에 결함이 발생했다. 1% 가
량의 형질 전환 배아는 배아발생 14.5일에 뇌 형성의 기형을 보였다. 두 번째로 EphrinA2의 과잉 발현은
EphA8 수용체의 tyrosine kinase domain의 활성을 더욱 증가시켰고, EphA8의 tyrosine 인산화는 E14.5일 된 배아
중뇌의 앞부분에서 추출한 세포에서 MAPK 활성을 증가시킴을 알 수 있었다. 세 번째로 세포 부착력 실험(cell
attachment assay)을 통해 EphrinA2의 중뇌 앞부분에서의 과잉 발현은 fibronectin에 대한 세포의 부착력 (cell
attachment)을 증가시킨다는 것을 발견할 수 있었다. 이러한 연구 결과들을 통해 Eph 수용체와 Ephrin리간드의 상반되는 발현은 중뇌(mesencephalon)의 형성에 중요하게 관여할 것으로 여겨진다.The Eph family is thought to exert its function through the complementary expression of receptors
and ligands. The dorsal mesencephalon appears to be segmented into two broad regions demarcated by the mutually
exclusive expression of EphA receptors and ephrinA ligands. In this study, we analyzed transgenic embryos expressing
ephrinA2 in the anterior region of the developing midbrain where the EphA8 receptor is expressed. First, 1% of
transgenic embryos showed cephalic neural tube closure defects. Second, it was confirmed that mis-expression of
ephrin-A2 in the anterior mesencephalon induced an increase in the EphA8 tyrosine kinase activity. Accordingly, an
increased MAPK activity was also detected in the anterior mesencephalon of E14.5 transgenic embryo. Third, cell
adhesion assay revealed that mis-expression of ephrinA2 promoted cell attachment to fibronectin. Taken together,
these findings suggest that co-expression of EphA receptors and ephrinA ligands significantly alter cell behaviors
including cell adhesion.
