저분자열충격단백질(small heat shock protein)의 분자적 샤프론(molecular chaprone) 기능은 잘 알려져
있다. 그러나 중추신경계통의 정상 발생과정에서 저분자열충격단백질의 발현과 분포에 대하여는 알려진 바가
거의 없다. 본 연구에서는 닭 배아의 소뇌에서 저분자열충격단백질의 일종인 HSP27과 αB-crystallin (αBC)의 발
현을 정량적, 공간적으로 관찰하여 발생과정 중에 발현하는 저분자열충격단백질의 역할을 추정하고자 immunoblot,
면역조직화학염색 및 이중면역형광염색을 시행하였다. Immunoblot 결과 HSP27과 αBC는 공통적으로 발생
14일에 처음 발현이 관찰되었고, 발생 18일에는 발현이 증가함을 확인하였다. 면역조직화학염색의 결과 αBC면
역세포는 발생 14일에 소뇌 백색질에서 약한 면역반응을 나타내었고, 발생 18일에는 백색질에서 강한 면역염색
성을 보였다. 일부의 세포는 과립층에서도 관찰되었다. Myelin-basic protein (MBP)과의 이중면역형광염색에서 소뇌의 백색질과 과립층에서 관찰된 αBC면역세포는 희소돌기아교세포로 확인되었다. HSP27면역반응은 발생 14
일에 처음 소뇌 백색질과 조롱박층에서 약하게 관찰되었다. 발생 18일에는 백색질, 소뇌핵 및 조롱박층에서 면
역반응이 뚜렷하게 관찰되었으며 과립층에서도 드물게 관찰되었다. αBC와의 이중면역형광염색에서 백색질과
과립층에서 관찰된 HSP27면역세포는 희소돌기아교세포로 확인되었다. HSP27과 αBC는 공통적으로 말이집을
형성하는 성숙 희소돌기아교세포에서 발현하였고, HSP27은 성숙 단계의 조롱박세포와 소뇌핵의 신경세포에서
발현하였다. 정상 발생 소뇌의 특정 신경세포와 신경아교세포에서 HSP27과 αBC가 특정 시기에 발현이 시작되
고 이 후 발현이 증가하는 것은 각 세포의 성숙단계에서 요구되는 생리적 스트레스에 대응하기 위한 세포 방어기전의 일부로 판단된다.It is well known that small heat shock proteins play a role as molecular chaperone. However, during
normal development of the cerebellum, expression and distribution of HSP27 and αB-crystallin (αBC) which are small
heat shock proteins have not been reported. To verify the protective role of HSP27 and αBC in neurons and glial cells,
we examined the expression and distribution of HSP27 and αBC in the developing chick cerebellum using immunoblot,
immunohistochemical and double immunofluorescence staining. Expression of both HSP27 and αBC was first
identified in the cerebellum of the embryonic day 14 (E14) embryo, and was increased at E18. Double immunofluorescence
analysis with myelin-basic protein (MBP) demonstrated that αBC positive (+) cells were mature myelinating
oligodendrocytes. αBC+ cells were observed in the white matter of the E14 cerebellum. At E18, there were a number
of αBC+ cells in the white matter and a few cells in the granular layer of the gray matter. On the other hand, HSP27+
cells were observed in the white matter and the Purkinje cell layer at E14. At E18, HSP27+ signals were observed in
Purkinje cells and neurons of cerebellar nucleus as well as oligodendrocytes in the white matter and the granular layer.
The results that HSP27 and αBC were expressed in specific neurons and glial cells in the developing cerebellum suggest
that HSP27 and αBC may be involved in the protective mechanism for the apoptosis of neurons and the physiological
stress occurred in oligodendrocyts during cell maturation.
