발생중인 실험동물 및 사람에서 고열은 선천성기형을 유발하는 요인의 하나로 인정되고 있다. 세포가
고열에 노출되면 방어기전으로 열충격단백질을 합성, 분비한다. Hsp70은 열충격단백질의 한 종류로서 정상발생
과정뿐만 아니라 고열과 같은 스트레스에 의한 세포자멸사를 억제하는 기능을 갖는 것으로 알려져 있다. 따라서
Hsp70이 결손된 실험동물이 고열에 노출되면 배아 혹은 태아에서 성장지연 및 선천성기형이 유발된다. 본 연구
에서는 임신한 Hsp70 KO 생쥐를 고열에 노출한 후 치아 초기발생과정에 중요한 역할을 수행하는 FGF-4와
FGF-8의 발현양상을 면역조직화학법으로 연구하고자 하였다. 실험군은 고열에 노출된 Hsp70 KO 생쥐 태아를,
대조군으로 고열에 노출되지 않은 Hsp70 WT 생쥐 태아를 사용하였다. 실험군은 임신 제8.5일에 43?C 수조에서
5분간 열충격을 주었고, 대조군에는 아무런 조작도 가하지 않았다. 임신 제13.5, 15.5 및 17.5일에 모체를 희생하
여 자궁내의 태아를 적출한 후 머리를 절취하였다. 머리에서 아래턱을 절취하여 통상적인 방법에 따라 파라핀에
포매한 후 조직표본을 제작하여 FGF-4 및 FGF-8 면역조직화학염색을 시행하고 광학현미경으로 관찰하여 다음
과 같은 결과를 얻었다. FGF-8 발현은 발생 13.5일 대조군에서는 치아판(dental lamina) 내부와 치아싹 첨부의
세포에서 양성반응을 보였고, 실험군에서는 극히 일부의 세포만 양성반응을 나타내었다. 발생 15.5일 및 17.5일
대조군에서는 치아속질(dental pulp)에 인접한 사기질모세포(ameloblast) 바닥막에서 양성반응을 나타내었으나,
발생 15.5일 실험군에서는 대부분의 치아상피에서 양성반응을 보였고, 발생 17.5일 실험군에서는 치아목고리 주
변의 바깥 및 속치아상피에서 강한 양성반응을 나타내는 등 발생이 진행되어도 지속적으로 발현하고 있었다.
FGF-4 발현은 발생 13.5일 대조군에서는 치아상피의 첨부에서 양성반응을 보였고, 발생 13.5일 실험군에서는 반
응이 관찰되지 않았다. 발생 15.5일 대조군에서는 치아상피의 바닥막과 치아목고리에 위치한 치아상피세포에서
양성반응을 나타내었으며, 실험군에서는 치아싹은 초기 모자시기 단계였고, 치아판 입구와 치아목고리에 위치한
치아상피에서 양성반응을 보였다. 발생 17.5일 대조군에서는 이차사기질결절로 분화할 부위에서 양성반응을 보
였고, 발생 17.5일 실험군에서는 구강상피에 인접한 치아판에서 강한 양성반응을 보였으나 나머지 부위에서는
미약하거나 음성반응을 나타내었다. 이상의 결과를 종합하면 임신 중 고열은 치아발생이 진행되어도 FGF-8을
지속적으로 발현시키고, 또한 FGF-4의 발현을 저해하여 치아의 발생과정을 억제하는 것으로 생각된다.To investigate the effects of maternal hyperthermia on early odontogenesis, pregnant Hsp70 knockout
and wild type mice at embryonic day (ED) 8.5 were immersed in a 43?C water bath until their core body temperature
reached that temperature, and then given a further 5min of hyperthermia. Untreated Hsp70 WT mice fetuses were
used as the control group. Fetuses were collected at EDs 13.5, 15.5 and 17.5. Developing teeth in the mandible were
processed for histological and immunohistochemical studies. Tissue sections were immunostained for FGF-8 and FGF
-4 and observed using light microscopy. In the controls, FGF-8 immunolocalization was observed in cells within the
dental lamina and in apically located dental epithelium at ED 13.5. However, a few cells were immunopositive in the
heat shocked (HS) group. At EDs 15.5 and 17.5 of the control group, the basal lamina adjacent to the dental pulp
showed positive immunostaining. In contrast, most of the dental epithelium was immunopositive at ED 15.5 in the HS
group and inner and outer dental epithelial cells were continuously immunopositive by ED 17.5. FGF-4 immunolocalization
was found in apical dental epithelium at ED 13.3 in the control group, but no such positive reaction was
observed in the HS group. At ED 15.5 in the controls, basal lamina and dental epithelium near the cervical loop were
immunopositive. In contrast, early cap-stage teeth had cells near the mouth of the dental bud and cervical loop that
were immunopositive to FGF-4 in the HS group. In controls at ED 17.5, cells near the future secondary enamel knot
were immunopositive, whereas most of the dental epithelium except for cells in the mouth of the dental lamina was
negative in the HS group. Thus, maternal hyperthermia may inhibit normal odontogenesis through sustained production
of FGF-8 and downregulation of FGF-4.
