Tissue inhibitor of metalloproteinases (TIMPs)는 TIMP-1, -2, -3, -4로 이루어진 단백질 그룹으로 대부
분의 기질단백분해효소의 작용을 억제한다. 기질단백분해효소의 활성은 사이토카인, 성장인자, 호르몬 등을 통한
발현 조절과 불활성 전구체의 활성화를 통해 조절되는데, 관절염, 동맥 경화, 염증 질환, 암의 성장 및 전이과정
등에서 기질단백분해효소와 TIMP 간의 균형에 이상이 초래된 것이 발견된다. 본 연구에서는 대뇌 허혈 후
TIMP-1의 발현 변화를 조사하고, 발현 조절에 관여하는 promoter 부위를 동정하고자 하였다. 1시간 동안 중대뇌
동맥을 폐쇄한 후 재관류시키는 국소허혈 동물 모델을 사용하여 TIMP-1의 발현을 조사한 경우, 재관류 1일과 2
일 후에 경색 주변부에서 발현이 현저히 증가한다. 재관류 1일과 2일의 경색 주변부에서 집중적으로 활성화되
는 것으로 알려진 별아교종양세포를 사용하여 TIMP-1이 발현을 조사한 결과, TIMP-1은 interferon gamma
(IFNγ)를 처리하여 활성화시킨 C6 별아교종양세포주에서 발현 증가를 보인다. Reporter gene을 사용하여 TIMP-
1 promoter의 IFNγ에 대한 반응을 조사한 결과, TIMP-1 promoter의 -226과 -115 사이에 존재하는 두 개의
SP1 결합 부위가 IFNγ에 의한 TIMP-1 발현 유도를 조절할 가능성이 있음을 보였다. 해당 promoter 부위에 존재
하는 SP1 결합 부위의 중요성을 조사하기 위하여, SP1 결합부위의 sequence 일부를 다른 sequence로 치환한 돌
연변이 reporter construct를 제작하여 C6 별아교종양세포주에 발현시킨 후 reporter gene의 발현을 조사한 결과,
IFNγ에 의한 TIMP-1 발현 유도가 관찰되지 않았다. 이와 같은 결과는, TIMP-1은 염증 자극에 의하여 별아교세
포에서 발현이 증가하며, 이와 같은 발현 증가에 TIMP-1의 promoter에 존재하는 SP1 결합 부위가 중요한 역할
을 할 가능성을 시사한다.Tissue inhibitors of metalloproteinases (TIMPs) comprise a family of secreted multifunctional
proteins that consists of four members (TIMP-1 to TIMP-4). TIMPs are all major inhibitors of most matrix
metalloproteinases (MMPs). They are synthesized by a variety of different cells and regulated by a number of
cytokines and by growth and differentiation factors. The balance between MMPs and TIMPs plays a crucial role in the
turnover of extracellular matrix in normal and pathological conditions. Here, we report that the production of TIMP-1
was upregulated in interferon (IFN)γ-treated C6 astroglioma cells and that the proximal 226 bp region of the promoter
of the TIMP-1 gene is responsible for IFNγ-induced induction in C6 astroglioma cells. The induction of TIMP-1
production by IFNγ was virtually abolished by introducing mutations into the putative SP1-response element in the
promoter, indicating that the SP1 binding site conferred responsiveness onto a heterologous promoter. Together the
results suggest that the IFNγ-induced upregulation of TIMP-1 production in C6 astroglioma cells is mediated by the
SP1 binding site localized in the TIMP-1 gene promoter.
