본 실험은 영구 중뇌동맥폐쇄(permanent middle cerebral artery occlusion) 후 electrogenic Na+/HCO3
-
cotransporter (eNBC)와 Na+/H+ exchanger 1 (NHE1)의 발현변화를 알아보기 위하여 시행하였다. 허혈주변지역에
서 eNBC와 NHE1의 발현은 3시간군과 6시간군에서 의미있게 증가하였다. 국소허혈 후 허혈주변지역에서의
eNBC와 NHE1 발현의 증가는 뇌세포의 알카리성화와 나트륨의 과다유입을 통해 뇌세포의 부종과 세포손상을
초래할 것으로 생각된다. 따라서 국소허혈 후 허혈주변지역에서 나타나는 eNBC와 NHE1의 증가는 이들 지역에
서의 이차적인 신경세포손상에 중요한 역할을 할 것으로 생각된다.We conducted a chronological and comparative analysis of the levels of the electrogenic Na+/HCO3
cotransporter (eNBC) and Na+/H+ exchanger 1 (NHE1) on ischemic penumbra, using the rat model of permanent
middle cerebral artery occlusion (pMCAO). Both eNBC and NHE1 levels were significantly higher in the ischemic
penumbra from 3 h to 6 h after focal ischemia than in the sham-operated control group. The enhancement of eNBC and
NHE1 production following focal ischemia may contribute to brain cell swelling or damage during ischemic penumbra
through intracellular alkalinization and Na+ overload. Therefore, increases of eNBC and NHE1 in ischemic penumbra
may play an important role in secondary brain cell damages following permanent focal ischemic insults.
