Vinblastine, an alkaloid isolated from periwinkle plant (Vinca rosea Linn.), is presently used extensively in cancer therapy. But it has been known to possess various biologic effects, such as depression of bone marrow, arrest of cell division in metaphase, etc. It also possesses the ability to bind to tubulin, the protein thought to be the major component of microtubules, and the loss of microtubules on the renal podocytes was reported.
In the present investigation vinblastine was administered intravenously in doses of 0. 2 and 0.3mg/kg to rabbits and the effects on the renal function were studied. And it was further attempted to elucidate the mechanism of the renal action of vinblastine, utilizing phentolamine, vasopressin and spironolactone.
The results were summarized as follows:
1. Vinblastine decreased the urine flow and urinary sodium excretion as well as renal plasma flow and glomerular filtration rate. Also, the free water reabsorption was markedly depressed by vinblastine. These responses increased roughly in proportion with the dose administered.
2. Systemic blood pressure was not affected by vinblastine.
3. The changes of various parameters of renal function by vinblastine significantly correlated one another, indicating that overall renal response to vinblastine was brought about mainly by the hemodynamic changes.
4. Pretreatment of phentolamine significantly inhibited the actions of vinblastine on the urine flow rate, renal plasma flow, glomerular filtration rate and urinary sodium excretion. However, the action on free water reabsorption was not affected by phentolamine.
5. Infusion of vasopressin significantly diminished but not abolished the decrease in free water reabsorption induced by vinblastine.
6. Spironolactone did not influence the actions of vinblastine.
From the above results, it was suggested that the antidiuretie effect of vinblastine in rabbit is caused primarily by the changes of renal hemodynamics, which, in turn, was induced by the sympathetic activation on the renal artery.*표시는 필수 입력사항입니다.
