약물 하나를 개발하기 위해서는 수 많은 시간과 비용이 발생한다. 하지만 많은 투자를 하고 있음에도 불구하고 약물 개발에 있어 성공률은 높지 않다. 이는 약물 개발 단계에서 임상시험 전에 전임상 시험 단계를 거치는데 이 때 후보약물 선별에 있어 판별 방법이 약물 개발의 진행을 늦추고 있다.
전임상 단계에서는 세포 배양과 동물실험이 진행되는데 세포배양은 플라스틱 디쉬에서 세포를 2차원으로 배양하여 약물에 대한 반응을 테스트한다. 하지만 이러한 방법은 실제 생체와 다른 환경을 나타내어 세포의 특성을 완전히 나타내지 못하며 또한, 세포-세포 간의 상호작용을 통한 약물반응을 알지 못한다. 그리고 후보약물을 판별하는 다른 방법은 동물시험이다. 동물실험은 2차원 세포배양보다는 장기-장기의 상호작용을 통한 반응을 알 수 있으나 사람과 다른 유전자로 인해 동물실험에서 통과된 약물을 임상 시험에서 적용할 때 많은 실패율을 나타내기도 한다. 이러한 문제점을 해결하기 위해서는 생체 내의 환경을 반영할 수 있는 환경과 인간 유도 만능 줄기세포와 같이 사람 세포를 반영할 수 있는 시스템에서 후보 약물 테스트가 필요하다.
오르간 온 어 칩은 마이크로 사이즈, 적은 투여 약물 용량, 오르간과 오르간 사이의 상호작용 등으로 인해 약물 개발 산업에서 약물을 개발하는데 있어 유망한 도구로 떠오르고 있다. 최근에 인간 유도 만능 줄기세포로 제작된 3 차원 배양기술과 함께 적용되어 후보 약물 판별에 있어 성공률을 높여줄 플랫폼이 활발하게 연구되고 있다. 본 연구에서는 세포-세포 상호작용을 할 수 있는 플랫폼 개발 및 다양한 마이크로티슈 제작과 특성 분석을 하였다.
첫번째 제안하는 플랫폼은 상용화된 96 웰 플레이트를 사용하여 물리적으로 분리된 웰들을 개발한 디바이스를 사용하여 서로 연결해주는 플랫폼이다. 즉, 각 웰에 있는 미디어를 디바이스를 통해 공유하여 공간적으로는 마이크로 오르간들이 떨어져 있지만 유체적으로는 연결되어 있는 플랫폼이다. 정체되어 있는 환경과 디바이스를 사용하여 흐름이 있는 환경에서 대장암 마이크로 티슈를 배양한 결과 흐름이 있는 환경에서의 암세포가 훨씬 크기가 커졌다. 또한 항암제를 (5-FU) 사용하여 약물스크리닝을 한 결과 농도에 따라 암세포의 크기가 달라지는 것을 확인하였다.
두번째 플랫폼은 96 웰 형태를 기초한 미세유체 디바이스이며 5개의 서로 다른 마이크로티슈를 (뇌 피질,해마, 심장, 간, 암) 디바이스 안에서 배양하여 제작한 배지와 원래의 배지를 사용하여 제작한 배지가 원래의 배지만큼 각 마이크로티슈들의 특성을 잘 나타냈음을 확인하였다. 제작한 배지를 사용하여 약물 5-FU 의 존재 유무에 따른 장기들의 반응과 항암효과를 분석하여 확인하였다.
세번째 플랫폼은 두번째 플랫폼의 단점을 보완한 96 웰 형태를 기초한 미세유체 디바이스이다. 제안된 디바이스에서도 암세포 마이크로티슈가 정적인 환경보다 디바이스를 사용한 흐름이 있는 환경에서 더 잘 배양되는 것을 확인하였다. 또한 농도별로 약물 스크리닝을 통해 결과를 통해 흐름이 있는 곳에서 더 항암효과가 있는 것을 확인하였으며 우리 신체처럼 흐름이 있는 환경에서 약물 스크리닝을 해야 함을 보여주었다.
요약하면, 상용화된 플레이트를 사용하여 손 쉽게 각 웰들을 연결할 수 있는 플랫폼과 96 웰 형태의 디바이스에서 6 개의 웰들이 미세유체채널을 통해 서로 상호작용할 수 있는 플랫폼을 개발하였다. 이 연구에서 개발한 플랫폼들은 약물 개발에 있어 전임상 연구와 더 복잡한 멀티 오르간 네트워크 시스템 플랫폼으로 후보약물 평가에 있어 시작점이 되질 바라며, 앞으로 새로운 바이러스가 발현되어 빠른 시일 내에 약물을 개발할 때 후보 약물 평가에서 비용과 시간을 줄 일 수 있도록 평가 방법에 기여하길 바란다.*표시는 필수 입력사항입니다.
