관절 연골의 세포외기질 (Extracellular matrix, ECM)은 연골 세포의 기능과 연골 조직의 항상성을 유지하는 데 중요한 역할을 한다. 그러나 연골은 혈관과 신경이 없어 연골 손상 시 조직 재생이 힘들고 조직재생을 위한 자가 연골세포이식술(ACI) 또한 이식에 필요한 연골 세포가 세포 증식 과정 중에 탈분화 (dedifferentiation of chondrocyte)가 되는 한계점을 가지고 있다. 연골 세포의 탈분화는 무릎에 가해지는 충격을 흡수할 수 있는 기계적 성질을 형성하는 제2형 콜라겐과 aggrecan 합성이 감소하고 연골 비특이적 ECM인 제1형 콜라겐이 증가하면서 섬유아세포와 같은 표현형을 가지게 된다. 연골 특이적 기질의 손실은 연골 조직 기능에 필요한 기계적 성질을 저하시키고 결과적으로 세포 이식 후 연골 재생과 회복이 지연된다. 따라서 연골 재생의 한계를 극복하기 위해서는 연골 세포 탈분화 기전을 이해하고 탈분화 기능제어의 연구가 수반되어야 한다. 이에 본 논문에서는 연골 세포 탈분화 과정에서 관찰되는 섬유아세포와 유사한 세포 모양 변화 (spindle cell shape)에 착안하여 세포-기질 상호작용이 매우 중요하게 작용할 것으로 가정하고 연구를 진행하였다. 이 중에서도 기질의 물리적 특성과 이를 세포 내 신호로 전달 전달하는 매개분자인 focal adhesion kinase (FAK)의 상호작용이 세포 내 견인력과 장력 형성에 미치는 영향과 이에 따른 탈분화 과정 중에 관찰되는 유전자의 발현 변화를 중점으로 연구를 진행하였다.
첫째, 연골 세포의 탈분화 동안 세포의 핵막 단백질인 lamin A의 발현 변화를 연구하였다. 탈분화 중 연골 세포의 Lamin A 발현 및 핵의 물리적 특성과 그에 따른 유전자 발현변화에 대한 연구가 완전히 조사되지 않았기에, chapter 1에서는 탈분화 중 관찰되는 cell spreading이 actin stress fiber형성을 통해 세포 견인력 및 장력을 형성하고 이는 Lamin A 발현 변화를 유도하여 핵의 구조적, 물리적 특성을 변화시킬 것으로 가정하고 실험을 진행하였다. 연구의 결과는 기질의 물리적 강도에 의해 증가된 세포 내 견인력과 장력은 Lamin A 단백질 발현을 증가시키고 동시에 핵의 장력 또한 증가함을 발견하였다. 또한, 세포와 상호작용하는 기질의 물리적 강도에 따라 Lamin A와 탈분화의 특징인 제1형 콜라겐과 Lox의 발현이 조절될 수 있음을 발견하였다. 이는 연골세포의 탈분화 과정에 수반되는 세포 내 장력과 Lamin A 발현이 탈분화의 표현형인 연골 비특이적 기질 합성을 조절할 수 있음과 동시에 탈분화 기능을 제어할 수 있는 가능성을 제시한다.
둘째, 기질의 물리적 강도 조절과 focal adhesion(FA) 억제 실험을 통해 세포-기질 상호 작용이 연골 세포 탈분화 표현형에 끼치는 영향을 연구하였다. 본 연구에서 세포 견인력 현미경(TFM-IFM)을 사용하여 물리적 강도가 약한 기질에서 배양되거나 FAK inhibitor (Y15) 또는 Lox inhibitor (BAPN)와 함께 배양된 연골 세포는 integrin-FA complex, actin cytoskeleton 형성과 제1형콜라겐과 Lox 생산이 저하되는 것을 보여주었다.
종합적으로, 본 학위논문에서는 Lamin A 의 연골 세포 탈분화의 새로운 분자 마커로써의 가능성과 잠재적인 치료 표적을 제공할 것을 기대함과 동시에, 본 논문에서 보여준 세포-기질 상호작용을 통한 연골 세포 탈분화 기능제어 또한 연골조직 재생공학을 위해 잠재적으로 이용될 수 있을 것으로 기대한다.*표시는 필수 입력사항입니다.
