카보닐화 반응은 단일탄소 화합물인 카보닐 분자를 화합물에 도입할 수 있는 매우 유용한 합성 방법으로 제약, 농업, 신소재 등 다양한 분야에서 활용되고 있다. C1 빌딩 블록인 일산화탄소(CO)는 대표적인 카보닐 소스이며, 높은 반응성과 활용성으로 여러 합성반응에 사용되고 있다. 또한, CCU 기술인 드라이 리포밍, RWGS 등 공정을 통해 CO₂ 로부터 전환된 CO를 활용함으로써 온실가스 배출을 줄이고 지속 가능한 화학 공정 개발에 기여할 수 있다. 이러한 CO를 도입하는 카보닐화 반응 중 산화카보닐화 반응은 산화제를 사용하여 CO를 도입하는 반응으로 팔라듐 촉매를 사용하는 다양한 반응이 개발되었다. 특히 CO와 산소를 활용하는 산화카보닐화를 통해 우레아, 카바메이트, 카보네이트 등을 생산하는 반응은 유독성 물질 사용을 대체하는 공정으로 활용할 수 있어 다양한 연구가 보고되고 있다.

CO, O₂, 알코올 그리고 다이아민을 사용하는 산화 카보닐화 반응은 디카바메이트를 합성할 수 있는 효과적인 방법이다. 디카바메이트는 열분해를 통해 디이소시아네이트를 용이하게 생산할 수 있다. 이러한 반응을 활용하면 유독성 물질인 포스겐을 사용하는 현재의 폴리우레탄 생산 공정을 보다 친환경적인 공정으로 대체할 수 있다. 또한 CO₂에서 유래한 CO를 사용하여 디이소시아네이트를 합성하면, 저탄소 폴리우레탄 생산도 가능하다.

산화 카보닐화는 단일 공정으로 아민기에서 카바메이트를 생성할 수 있는 효과적인 방법임에도 불구하고, 오래도록 모노아민 카보닐화 반응 연구만 다수 보고되었고, 다이아민의 카보닐화 반응 연구가 정체되어 있었다. 이는 카보닐화 반응에 의해 두개의 아민이 결합하는 반응 매커니즘 때문에 다이아민을 반응물로 사용할 경우, 카바메이트가 생성되기 전에 부산물인 우레아 폴리머가 생성되기 때문이었다. 따라서 산화 카보닐화를 통한 디카바메이트 합성연구는 보고된 사례가 드물었고, 특히 재사용이 가능한 불균일계 촉매를 사용한 디카바메이트 합성 연구는 거의 없다.

최근 Leitner 등이 발표한 논문은 유일하게 불균일계 촉매를 사용하여 디카바메이트를 합성한 사례인데, 해당 논문에서도 역시 우레아 폴리머 생성을 디카바메이트 합성 수율 저하의 원인으로 지적했다.

본 연구는 아민 산화 카보닐화 반응의 매커니즘과 반응 특성을 살펴보고 다이아민 산화카보닐화와 모노아민 산화 카보닐화의 차이를 살펴보았다. 또한 앞서 살펴본 매커니즘 연구를 통해 카바메이트가 합성되기까지 산화카보닐화와 메톡시화가 순차적으로 발생하며, 반응 중간체로 우레아가 발생하는 것을 확인할 수 있었다.

이러한 매커니즘 연구를 기반으로 산화카보닐화와 메톡시화의 속도 차이를 줄일 수 있는 불균일계 촉매 Pd/CeO₂를 개발하였다. 또한 Pd/CeO₂가 메탄올을 메톡시 상태로 쉽게 활성화시키며 안정적으로 유지하는 특성을 밝혔다. 이러한 특성으로 인해 우레아 중간체를 빠르게 디카바메이트로 전환할 수 있고 산화 카보닐화와 메톡시화의 속도 차이를 줄여 디카바메이트 수율이 향상되는 것을 확인하였다.

카바메이트 합성 과정이 아민 카보닐화를 통한 우레아 중간체 생성, 그리고 우레아 중간체의 메톡시화 반응을 통한 카바메이트 생성이라는 것이 이전 연구를 통해 알려져 있는데, 디카바메이트 합성의 경우 다이아민의 두 개의 아민기가 모두 위와 같은 과정을 거쳐야 디카바메이트가 생성된다. 이 때 카보닐화는 낮은 온도에서도 빠른 속도로 진행되는 반면, 메톡시화 반응은 높은 온도 조건에서만 효과적으로 진행되는 것을 확인했다. 따라서 메톡시화 진행이 어려운 승온과정(저온조건)에서 카보닐화 반응만 발생하는 것이 우레아 폴리머 생성의 원인이었다. 이러한 문제를 해결하기 위해 산화 카보닐화 반응에 필수적인 산화제 투입 시기를 조절하여 효과적으로 우레아 생성을 막을 수 있었으며, 디카바메이트 합성 수율이 향상되었다.

앞서 밝힌 것과 같이 우레아 폴리머 발생은 다이아민 산화 카보닐화를 통한 디카바메이트 합성에 주요한 수율 저하의 요인이다. 새로운 우레아 폴리머 생성 억제 방법으로 모노 아민을 활용했다. 연속된 산화 카보닐화 반응으로 분자량이 커지면 우레아 중간체는 쉽게 반응성을 잃고 메톡시화에 참여하지 않는다. 따라서 우레아 폴리머 생성을 억제하기 위해 모노 아민을 반응 제제 사용하였고, 모노 아민이 효과적으로 작용하는 것을 알 수 있었다. 또한 모노아민의 카보닐화, 메톡시화 속도를 비교하여 m-anisidine이 반응 억제제로서 효과적으로 작용하는 이유를 규명하였다.
