식물 생명공학 분야에서 유전물질을 세포 내로 정확하고 효율적으로 전달하는 기술은 신속한 품종 개량 및 기능 유전자 분석을 위한 핵심 요소로 부각되고 있다. 그러나 기존의 대표적인 방법인 Agrobacterium 매개 유전자 도입 방식은 일부 모델 식물이나 특정 작물에만 적용이 가능하고, 유전체 내 무작위 삽입으로 인해 의도하지 않은 변화를 유발할 수 있다는 한계가 존재한다. 이러한 기술적 제약은 식물 세포 특성에 적합한 전달체 개발, 고효율 유전체 교정 시스템 확립, 그리고 안정적인 식물세포 재분화 플랫폼의 구축이라는 세 가지 측면에서 동시에 해결되어야 한다.본 논문에서는 이러한 한계점들을 극복하기 위해 나노 소재 기반의 유전물질 전달체 개발, 높은 교정 효율을 가진 염기 및 프라임 에디터의 선별, 원형질체 유래 식물체 재분화 시스템의 최적화에 대한 연구를 진행하였다.
식물세포 내부로 외부 유전물질을 효율적으로 전달할 수 있는 나노 소재 기반의 연구를 1장에서 진행하였다. 단일벽 탄소나노튜브 (Single-walled carbon nanotube; SWNT)와 지질 나노입자 (Lipid Nanoparticle; LNP)를 전달체 후보군으로 선정하였다. 첫째로, 유전물질을 식물 세포질에서 방출할 수 있게 SWNT 표면을 Py-His6 그룹으로 개질시킨 pH 민감성 나노입자 운반체 (His6-SWNT)를 개발하였다. STTM396-SWNT 복합체가 처리된 애기장대와 토마토 캘러스에서 증진된 재분화율을 관찰하였고, 이는 실제 캘러스 내 miR396 발현 수준 억제를 통해 식물 재분화 증진 효과로 이어짐을 확인하였다. 두번째로, MC3 기반 LNP (N/P = 5)가 독성이 낮고 높은 전달 효율을 보이는 것을 확인하였고, PBS (pH7.4) 기반으로 제조된 MC3-LNP를 5 mg/L로 처리 한 후 24시간 동안 식물 세포에 처리 후 pH 7.4에서 배양하였을 때 최대 전달효율을 확인하였다.
2장에서는, 크리스퍼 기술 기반의 염기교정 (Base editing) 및 프라임 에디팅 (Prime editing) 기술의 에디터 선정을 위한 연구를 진행하였다. 첫째로, 아데닌 염기 교정 기술 (Adenine base editor; ABE) 관련 에디터 5종류, 시토신 염기 교정 기술 (Cytosine base editor; CBE) 관련 5종류의 에디터 후보군에 대해 애기장대와 콩 원형질체에서 스크리닝을 수행하였다. 타겟 서열의 위치별 교정효율 (Positional editing efficiency), 각 에디터들의 교정 가능 영역 (Editing window), 그리고 교정 순도 (Product prutiy)에 대한 비교 분석을 통해서 가장 높은 활성을 가진 ABE8e (ABE)와 A3A/Y130F-V04 (CBE)를 선별하였다. 두번째로, 모든 경우의 염기 간 치환 외에도 삽입, 결손 돌연변이를 가능하게 하는 프라임 에디팅 기술 (Prime editor; PE)에서의 높은 교정 활성을 가진 에디터의 선별 및 개발에 대한 연구를 수행하였다. 총 8종류의 PE 후보군을 애기장대 원형질체 기반의 스크리닝을 수행한 결과, 가장 높은 활성을 가진 v4e2를 선별하였다.
마지막으로 3장에서는, 효율적인 원형질체 재분화 시스템이 확립되지 않은 식물 종에서의 최적화 및 플랫폼 개발 연구를 진행하였다. 첫째로 애기장대 식물체로부터 분열능이 있는 원형질체를 분리하기 위한 조건을 최적화하였다. 알긴산 하이드로젤 (Alginate hydrogel)을 기반으로 하여 안정적으로 세포 배양을 위한 조건을 확립하였고 이후 배양과정인, 원형질체 분열 유도 - 캘러스 형성 - 지상부 조직 재분화 – 뿌리 조직 분화 유도를 통한 식물체 생산, 각각에 대한 최적화된 배지 및 배양 조건을 확보하였다. 4종류의 애기장대 야생종 (ecotype)간 비교실험을 통해 재분화율이 높은 Ws-2 (Wassilewskija-2) 야생종을 선별하였으며, 3종류의 재분화 된 지상부 식물체 절편 (Shoot explant)에 대해 뿌리 조직 재분화율을 비교한 결과, 개화기 줄기 (inflorescence stem)가 뿌리 기관형성 (Root organogenesis)에 높은 효율을 보이는 것을 확인하였다. 두번째로, 원형질체 재분화 시스템이 확립되지 않은 토마토 품종인 마이크로톰 (Micro-Tom)에서 분열능이 있는 원형질체 분리, 배양 배지 및 조건의 최적화 과정을 거쳐 alginate hydrogel 기반의 안정적이고 효율적인 재분화 시스템을 개발하였다.
종합적으로, 유전물질 전달체 개발 연구, 높은 교정 활성을 가진 에디터 선정 연구, 그리고 안정적인 원형질체 유래 식물체 재분화 시스템 개발 연구는 다양한 식물종의 유전체를 교정하여 원하는 특성을 가진 품종을 빠르고 효율적으로 개발할 수 있는 플랫폼 구축에 기여할 것으로 기대한다.*표시는 필수 입력사항입니다.
