 본 연구는 천연 유화제 개발을 위한 일환으로, 우유 유래 극성 지질(Milk Polar Lipid, MPL)을 Phospholipase A2(PLA₂)로 가수분해한 h-MPL(hydrolyzed MPL)의 다양한 pH 조건에서의 유화 특성과 안정성, 그리고 아이스크림의 적용 가능성을 평가하였다. MPL에 PLA₂를 처리한 결과, 유리지방산의 방출과 C=O ester 피크의 감소가 FT-IR 분석을 통해 확인되었으며, 이는 PLA2에 의하여 MPL이 효율적으로 가수분해되었음을 의미한다. 이후, 합성 유화제인 Diacetyl Tartaric Acid Ester of Mono- and Diglycerides (DATEM), MPL과 h-MPL을 각각 우유 단백질이 있는 유제품 emulsion에 첨가하여 다양한 pH(3, 5, 7)로 제조되었으며, 이는 평균 지방구 부피 직경, 계면 단백질 농도 및 조성, 점도, 그리고 일주일간 저장 후의 크리밍 및 상분리 현상을 통해 유화 특성이 평가되었다. pH 7에서는 h-MPL 첨가군이 대조군과 MPL 첨가군에 비해 유의적으로 높은 계면 단백질 탈착률을 보였고, 이로 인해 지방구 크기가 감소하며 크리밍 안정성이 향상되었다. pH 3의 산성 조건에서는 h-MPL이 모든 처리구 중 가장 우수한 유화 특성과 저장 안정성을 나타냈으며, 이는 형광감도 분석, 표면소수성 그리고 FT-IR을 통하여 단백질과 극성 지질의 상호작용으로 인한 것임이 확인되었다. 우유 단백질과 h-MPL 간의 상호작용을 통해 친수성이 증가한 복합체가 형성되었기 때문으로 분석되었다. 형광 스펙트럼 분석 결과, h-MPL 처리 시 단백질 구조가 변형되어 형광 감도가 감소하였고, 이는 내부 소수성 잔기가 극성 환경으로 이동했음을 시사한다. 또한, 표면 소수성 측정 결과에서도 h-MPL 복합체는 MPL보다 낮은 값을 나타내었으며, FT-IR 분석을 통해 amide Ⅰ 및 Ⅱ 피크의 이동올 통하여 이는 단백질 2차 구조의 변형되었음을 확인하였다. 이는 MPL보다 h-MPL이 친수성 복합체를 형성하여 단백질 간의 소수성 상호작용으로 인한 응집이 억제되었음을 보여주었다. 반면, pH 5에서는 우유 단백질의 등전점에 근접함에 따라 정전기 반발력이 약화되고, 단백질 간 응집이 증가하여 입자 크기 증가, 점도 상승, 크리밍 가속 등의 유화 안정성 저하 현상이 모든 처리군에서 관찰되었다. 아이스크림 적용 실험에서는 h-MPL이 MPL 대비 유의하게 높은 오버런과 지방구 불안정화를 유도하였고, 단백질 탈착 및 미세구조 측면에서도 긍정적인 효과를 보였다. 또한 DATEM과 비교하였을 때, 아이스크림의 녹는 속도에 부정적인 영향을 미치지 않아, 제품 품질을 유지하면서 천연 유화제로 기능할 수 있음을 확인하였다. 이와 같은 결과는 h-MPL이 합성 유화제를 대체할 수 있는 천연 유화제로서 높은 기능성과 적용 가능성을 지님을 시사하며, 다양한 식품 환경에서도 유효한 친환경 대체 소재로 활용될 수 있는 잠재력을 제시한다.
