Polycaprolactone (PCL) cylindrical scaffolds with gradually increasing pore sizes along the longitudinal direction were fabricated by a centrifugation method to evaluate the effect of pore size on the differentiation of adipose-derived stem cells (ASCs) into osteoblasts (osteogenesis) and myoblasts (myogenesis). The prepared cylindrical scaffold showed gradually increasing pore sizes (from ~90 to ~400 μm) along the cylindrical axis. From the in vitro culture of ASCs, the different pore sizes in the scaffold were shown to provide different optimum environments for ASC differentiations (i.e., 300~320 μm for osteogenesis and 90~105 μm for myogenesis in our system). The pore size gradient scaffolds fabricated by the centrifugation method may be a good tool for basic studies, including determining the adequate ranges of pore size for a variety of stem cell differentiation into specific cell types.
