In this study, recombinant human growth hormone(rhGH)-loaded liposomes having high loading efficiency and stability were prepared. Loading efficiency of rhGH as a model drug was enhanced by applying modified freeze-thaw method at reduced pressure. The liposomes were prepared with a variation of total lipid amount and ratio in lipid composition. RhGH-loded liposomes had approximately 0.8~1.7 μm of mean particle diameter according to lipid composition and -1.0 ~ -10.0 mV of surface charge due to the presence of DSPE-mPEG 2000. Loading efficiency of rhGH was over 85% at an optimized lipid ratio. As the results of in vitro release test, rhGH-loaded liposomes showed a sustained release profile at 30:5.5:2.3 μmol ratio of HSPC:CHOL:DSPE-mPEG 2000 with relatively low initial burst release. Specific binding activity of rhGH assessed by ELISA assay was retained before and after manufacturing process of the liposomes and also during in vitro release test period of 72 hrs. Mean particle diameter and loading efficiency of rhGH-loaded liposomes rehydrated after freeze drying were retained especially when sucrose was used as a cryoprotectant. These results suggest that rhGH-loaded liposomes prepared by modified freeze-thaw technique may be applicable to rhGH delivery carrier for high loading efficiency, sustained release and stability with specific binding activity of rhGH.
